Core Lab Equipment: ABI 3100 Genetic Analyzer


Description:

The ABI 3100 Genetic Analyzer is used for fragment analysis and standard sequencing usually with a 50cm array with polymer 6 (Pop6), which gives sequence reads up to about 500-700 base pairs. Longer reads are run either on the 3130xl with Pop7 (up to 900bps) or on the 80cm array (800-1000 bps) which takes twice as much time and polymer.

The ABI PRISM Data Collection Software V.2.0 collects data in .FSA file format. Then ABI DNA Sequencing Analysis v5.2 and KB Basecaller V.1.1.1 are used to generate sequence files in .ab1 format. On request we can also generate FASTA, Standard Chromatogram (.scf) or Phred (.phd.1) files. Data files can be picked up in person with a jump drive, CD, or Zip disk or they can be posted to the Core web page and are password protected; please see Carrie Topp, in the Core Lab, for a username and password. Files can be further analyzed by software available for use in the Core Lab.
